Brief Description & Uses
Contains 30 or 60 dipsticks and necessary components to quantify the
amount of Apoptosis-Inducing Factor (AIF) from human sample material.
The kit includes sufficient materials to generate a standard curve and
prepare samples for analysis.
AIF is a flavoprotein (67 kDa) localized in the inter-membrane
space. It is a multi-functional protein with a vital oxidoreductase
function (required for cell survival), an anti-oxidant activity, an
assembly function in Complex I (NADH-ubiquinone oxidoreductase complex)
formation and an apoptogenic function. AIF has a major role in the
caspase-independent apoptosis process. It translocates to the nucleus
after mitochondrial release, leading to chromatin condensation and DNA
fragmentation.
The process of apoptosis, the main type of programmed cell death,
is an important function in cell termination, cell homeostasis, tissue
development and lymphocyte interactions.

Immunochromatographic Reader
While visual measurements and scanning on a flatbed scanner can both be
used to evaluate our dipsticks, the most effective quantitative results
are derived when using an immunochromatographic reader that has been
specifically designed to read conjugated gold lateral flow devices.
The reader that we recommend is the Hamamatsu ICA-1000,
which is a simple PC-based system for measuring and analyzing our
dipstick products. We have an agreement in place with Hamamatsu that
allows us to offer this reader directly to our customers. To learn more

The above results show the accuracy of the AIF dipstick kit (MSA31)
when measuring reduced amounts of AIF when a sample is diluted in 50%
increments. HepG2 cell extract was diluted 1:2 and 1:4 and equal
volumes of extract were loaded per dipstick. Graphical analysis was
performed using values interpolated from a standard curve. All analysis
was done with the MS1000 dipstick reader and GraphPad software.
