Brief Description & Uses

Contains a cytochrome c monoclonal antibody (50 µg) and an isotype-specific secondary antibody (50 µg) which has a fluorescein fluorophore conjugated to it.

Apoptotic cells which have released mitochondrial cytochrome c into the cytosol can be differentiated from non apoptotic cells which still retain cytochrome c in their mitochondria by fluorescence microscopy. The kit also contains an anti-Complex V α monoclonal antibody (50 µg) and the appropriate isotype-specific secondary antibody (50 µg) which has a Texas Red fluorophore conjugated to it. Complex V is an inner mitochondrial membrane protein and serves as a mitochondrial marker since it is not released into the cytoplasm during apoptosis, unlike cytochrome c. Thus one is able to observe cytochrome c and a mitochondrial marker (Complex Vα) simultaneously by fluorescence microscopy using MSA07. Buffers and blocking solution for immunocytochemical analysis are also included in the kit.

Purity: Near homogeneity as judged by SDS-PAGE. The antibodies were produced in vitro using hybridomas grown in serum-free medium, and then purified by biochemical fractionation.
Suggested Working Concentration: 2 µg/ml anti-Cytochrome C mab; 2 µg/ml anti-Complex V α mab;
2 µg/ml anti-mouse IgG2a-FITC; 2 µg/ml anti-mouse IgG2b-TXRD
Applications: Immunocytochemistry analysis of cytochrome c and a mitochondrial marker in apoptotic and non-apoptotic cells.
Antigen Species Cross-Reactivity: human, bovine, rat, mouse and C. elegans
Storage: The two monoclonal antibodies should be stored at 4°C. The two fluorophore-conjugated secondary antibodies should be stored, away from light, at 4°C. The goat serum should be stored at -20°C. The other components in the kit can be stored at room temperature.
