Brief Description & Uses
100 µg of monoclonal antibody reactive against the 33 kDa complex IV
assembly factor SURF1. Cellular levels of SURF1 are highest in growing
cultured cells.
SURF1 is an important assembly factor for cytochrome c oxidase
which is thought to participate in the folding of subunit II and
parallel Cu2+
insertion into this subunit. Mutations of SURF1, particularly ones that
result in an absence or near absence of the polypeptide, are the
leading cause of cytochrome c oxidase deficiency.
The anti-SURF1 mAb reacts specifically in human and bovine tissues.
The protein is easily detected in cells rapidly dividing their
mitochondria and has the highest levels in cells impaired in COX
assembly. The levels of SURF1 are always low and in some cases
neglible, even in rapidly dividing fibroblast cell cultures. Levels are
lowest in mature tissues such as heart or liver; the levels of SURF1
are in fact so low that a large amount of tissue material must be used
that the specificity is not as good as in cells. The low copy number is
consistent with it acting to catalyze Complex IV assembly. In cells
where cytochrome c oxidase assembly is incomplete because of mutations
in genes for structural subunits of the enzyme SURF1 is significantly
over-expressed. This is evident in Rho0 fibroblasts which are missing mtDNA and hence cannot synthesize subunits I, II and III of cytochrome c oxidase.
