Structure and Function: NDUFS4 is an 18-kDa subunit of Complex I, a 45-subunit complex of the respiratory chain. The NDUFS4 subunit makes up part of the iron-sulfur (IP) fragment of the protein. It has been claimed that the NDUFS4 subunit is phosphorylated in a cAMP dependent pathway that controls the activity of complex I. Mutations in this subunit, most often leading to loss of synthesis of the polypeptide, have been related to Complex I dysfunction in a number of patients with early onset mitochondrial diseases. It is thought that the subunit does not contribute to catalytic functions of Complex I. Useful in studies of Complex I assembly and Complex I deficiency in patients.
