Product Description
Provided is 50 µg of active, purified, recombinant human PDH Phosphatase 1 (PDP1). His-tagged PDP1 was bacterial-expressed and column-purified.
The enzyme is provided in a buffer perfectly suited for use with our PDH Enzyme Activity Assay Kits (MSP18 Microplate and MSP30 Dipstick).
Also available are PDH Protein Quantity Assay Kits (MSP19 Microplate and MSP31 Dipstick) for measuring total PDH levels, and companion phospho-specific detector antibodies against all three phosphorylation sites (also reactive in Western blotting).
Background
PDH kinases (PDK's) deactivate PDH by reversible phosphorylation. In humans and rodents, 4 PDK's are present in an organ-dependent manner. In all cases, PDK's are physically part of the PDH complex (binding via lipoyl domain 2 of E2), and they phosphorylate the E1 subunit at 3 specific serine residues. PDH phosphatases remove the phosphorylation and restore activity of PDH.
Our PDH Playbook provides an overview of the complex, along with descriptions of available experimental approaches for its analysis.
Our PDH Protocols provide very detailed instructions in the use of our PDH kinases and phosphatases with our activity assay kits, sandwich ELISA kits, and phospho-PDH antibodies.
